を添 加し、さらに 21 時間培養した。細胞抽出液の調製:トランスフェクション終了後の細胞を PBS (-) 400 L で

回洗浄後、PBS (-) で 5 倍希釈した細胞溶解剤 300 L を添加し、15 分間室 温でインキュベートし、さらに凍結(-80ºC)
および融解(37ºC)
を3
回繰り返 した。得られた細胞溶解液を 10,000 rpm で 10 分間遠心分離し、その上清を細 胞抽出液とした。ルシフェラーゼ活性の測定:細胞抽出液 20 L をルミノメーター用試験管
(Röhren Tubes、SARSTEDT)
に採取し、これに Luciferase Assay Substrate 100 L を添加した。 ルミノメーター (EG&G BERHTOLD 製 Lumat LB 9507) で 10 秒間の発光量を測定し、レニラルシフェラーゼ活性を評価した。細胞抽出液中のタンパク質の定量:細胞抽出液 10 L に BCA Protein Assay
Kit
溶液を 200 L 添加し、37ºC、30 分間インキュベートした。 その後、マイ クロプレートリーダー (Bio-Rad 製 Model 550) にて 570 nm における吸光度 を測定した。 細胞溶解剤でBSA
を希釈したものを標準液に用いて検量線を作 成し、上記ルシフェラーゼ活性およびタンパク質濃度より相対発光量 (RLU) を 算出した。107
件でルシフェラーゼ活性およびタンパク質濃度を求め、RLU を算出した。
