5‑2・3. カノレス形成および植物体の再生
改良伯州、15号 ' と せ き ほ く ' の プ ロ ト プ ラ ス ト 融 合 で は 249個 の コ ロ ニ ー が , 他 方 の 早 ど り 伯 州 ' と せ き ほ く ' で は 168個 の コ ロ ニ ー が そ れ ぞ れ 得 ら れ た . こ れ ら は 引 続 く 2週 間 の 培 養 で , そ れ ぞ れ 196個と 129個の 2mm大以上のカルスへと発達した.カノレスを再 分化培地に移植した結果, 改良イ白州 5号'の場合では 19儒 体 (9.7%) の カ ル ス か ら , 一 方 , 早 ど り 伯 州 ' で は 14個 (10.9%)のカノレスか ら 植 物 体 が 再 生 し た (Table5・2.,Fig.5・3.).
以 上 の よ う に , プ ロ ト プ ラ ス ト 融 合 試 験 で 合 計 33個 体 の 植 物 が 4 ヶ 月 間 の 再 分 化 試 験 中 に 獲 得 さ れ た が , 約 半 数 の 個 体 は 約 1年 の 培 養 期 間 で 維 持 増 殖 が 不 可 能 で あ っ た . す な わ ち , 発 根 不 良 , 葉 身 の ね じ れ , お よ び 生 育 遅 延 な ど の 種 々 の 奇 形 が 観 察 さ れ た . そ の 中 で 旺 盛 な 生 育 を 示 し , か っ , 充 分 な 発 根 が 認 め ら れ た HS‑8 と KS‑1の 2個 体 は , ガ ラ ス 温 室 内 に 設 置 し た ポ ッ ト で の 栽 培 が 可 能 で あ っ た .
HS‑8は 形 態 的 に は ネ ギ の そ れ と 類 似 す る が , そ の 葉 を 水 平 に 切 除 し た 断 面 は 半 円 型 で あ り , ネ ギ の 円 型 あ る い は タ マ ネ ギ の 三 角 型 と は 異 な っ た (Fig.5・4.).一方の KS‑1は,ネギ裂の円型の葉身を有するが,
地 下 部 に は 約 4cm径 の 球 根 の 形 成 が 観 察 さ れ た .
5・2・4. 細 胞 遺 伝 学 的 解 析
融 合 親 と 再 分 化 植 物 体 12個 体 に つ い て , フ ロ ー サ イ ト メ タ ー に よ り 核 DNA量 の 分 析 を 行 っ た . そ の 分 析 結 果 を Fig.5・5.お よ び Fig.5‑6. に 示 し た . ネ ギ の 最 初 に 出 現 す る ピ ー ク を 横 軸 の 50の 値 に 設 定 す る
と , タ マ ネ ギ は 50‑60の 値 で 最 初 の ピ ー ク ( 第 1ピ ー ク ) が 検 出 さ れた.ネギ 改良イ白州 5号 ' と タ マ ネ ギ せ き ほ く ' と の 組 合 わ せ を み る と (Fig. 5・5),KS‑6お よ び KS‑7は 50前 後 に 第 1ピ ー ク が 検 出 された.ただし, KS‑7は第 2番 目 の ピ ー ク ( 第 2ピ ー ク ) は 第 1ピ
