ネ ギ の 懸 濁 細 胞 培 養 を 確 立 す る た め に は 1mm径 の ス テ ン レ ス 製 メ ッ シ ュ 上 で 細 胞 塊 を 細 粒 化 す る こ と が 必 須 で あ っ た . 懸 濁 細 胞 培 養 の た め の 培 地 は BDS培 地 に 200mg • 1‑1の カ ザ ミ ノ 酸 を 添 加 し た 培 地 が 適 し て い た . ま た ,0.2Mの シ ョ 糖 を 添 加 す る こ と に よ っ て 細 胞 質 に 富 み 液 胞 化 し て い な い 細 胞 の 割 合 が 多 く な る こ と が 認 め ら れ た . カ ノ レ ス か ら の 構 物 体 再 生 率 は 0.5μMα‑naphtha1eneaceticacid (NAA)と1.0 μ M BAPの 添 加 し た 場 合 に 最 高 値 を 示 し た . ま た , カ ザ ミ ノ 酸 の 添 加 は 植 物 体 の 再 生 に 効 果 的 で あ っ た . カ ル ス か ら の 植 物 体 再 生 は , 継 代 培 養 回 数 が 7回 で は 約 70% で あ っ た が , 14代 の 継 代 培 養 回 数 で は 植 物 体 の 再 生 は 認 め ら れ な か っ た .
2ネ ギ の プ ロ ト プ ラ ス ト 培 養 お よ び 植 物 体 再 生 法 の 確 立
プ ロ ト プ ラ ス ト の 単 離 と 培 養 の た め の 諸 条 件 を 検 討 し た . プ ロ ト プ ラ ス ト の 単 離 は 酵 素 液 中 の マ ニ ト ー ル 濃 度 を 0.6M に し た 場 合 に 9.6X 106個・19FW‑1 と 最 高 値 を 示 し , フ ル オ レ セ イ ン ニ 酢 酸 染 色 に よ る 生 存 率 (Larkin,1976) は 95% 以 上 で あ っ た . プ ロ ト プ ラ ス と の 単 離 と 精 製 に 際 し て は 60Xgの 遠 心 分 離 条 件 が 適 当 で あ っ た .
BDS無 機 塩 組 成 を 基 本 に し て プ ロ ト プ ラ ス ト 培 養 に 適 し た 培 地 組 成 を 検 討 し た . そ の 結 果 , 硝 酸 カ リ ウ ム は 5m Mが 適 当 で あ り , 植 物 ホ ル モ ン は 2μM2,4・D と 1μMBAPの 組 み 合 わ せ で 細 胞 分 裂 率 が 最 高 値 を 示 し た . プ ロ ト プ ラ ス ト 培 養 の た め の 浸 透 圧 は 0.60osmol‑kg‑1
前後が適当であり, 0.2 M のショ糖, 0.2 M のブドウ糖,および 0.05M の 果 糖 の 添 加 で 細 胞 分 裂 が 促 進 さ れ た . 3種 の ビ タ ミ ン 組 成 を 検 討 し た 結果, TM‑2 (Shahin, 1985)が 最 適 で あ っ た . さ ら に , イ ノ シ ト ー ル , ア ス コ ル ビ ン 駿 お よ び シ ス テ イ ン の 適 正 濃 度 は そ れ ぞ れ , 0.56 m M, 1 m M, お よ び 1mMで あ る こ と が 示 さ れ た . ま た , プ ロ ト プ ラ ス ト 培 養 に 際 し て は 照 明 は 阻 害 的 で あ る こ と が 明 ら か と な っ た .
プ ロ ト プ ラ ス ト は 培 地 中 の 糖 濃 度 を 漸 減 す る こ と に よ っ て コ ロ ニ ー へ と発達した. 45日 目 の 小 カ ル ス を カ ル ス 形 成 培 地 に 移 描 し て そ の 発 達

を促した. 2mm大に発達したプロトプラスト由来カノレスを供試し,改 変 MS培 地 を 用 い て 植 物 体 再 生 試 験 を 行 っ た . 2 ‑ 3ヶ 月 後 に は 置 床 し た 75個 の カ ル ス の 中 の 25個 (32% ) か ら シ ュ ー ト の 形 成 が 認 め ら れ た . こ れ ら の シ ュ ー ト は 植 物 ホ ル モ ン を 含 ま な い 培 地 に 移 植 す る と 発 根 し た . さ ら に , こ れ ら の 幼 植 物 は ガ ラ ス 温 室 内 で 願 化 し た 結 果 , 正 常 に 生育した.
3 ネギとタマネギとの電気的細胞融合条件
プ ロ ト プ ラ ス ト 融 合 に よ り ネ ギ と タ マ ネ ギ と の 体 細 胞 雑 種 を 開 発 す る た め に 効 率 的 で 再 現 性 の あ る 電 気 的 細 胞 融 合 条 件 を 検 索 し た . 細 胞 融 合 液,電気融合のためのパラメーター,および綿胞密度の諸条件を検索し,
そ の 適 正 な 条 件 を 明 ら か に し た . ネ ギ は 懸 濁 培 養 細 胞 由 来 の 白 色 プ ロ ト プ ラ ス ト を 用 い , 一 方 , タ マ ネ ギ は 幼 植 物 自 来 の 葉 緑 体 を 有 す る 緑 色 プ ロ ト プ ラ ス ト を 単 離 し て , 両 者 の 細 胞 融 合 試 験 を 行 っ た . こ の 方 法 に よ っ て ヘ テ ロ カ リ オ ン を 容 易 に 識 別 す る こ と が で き た . 電 気 細 胞 融 合 の た め の 細 胞 懸 濁 液 は 0.5mMの カ ル シ ウ ム 溶 液 が 適 し て い た . プ ロ ト プ ラ スト融合のための電気パラメーターは, 1. 5 k V ‑cm ‑1 のパルス電界強度,
40秒の初期交流時間, 50μ 秒 の パ ル ス 揺 が 最 適 と 考 え ら れ た .1.5 X 105 個‑ml‑1の プ ロ ト プ ラ ス ト 密 度 で 融 合 を 行 う と 最 高 の ヘ テ ロ カ リ オ ン 形 成 率 が 得 ら れ , そ の 値 は 7 %以上であった.
4 電気融合によるネギとタマネギとの体細胞雑種の開発
電 気 細 胞 融 合 に よ り ネ ギ (Alliumfistulosum L., 2 n

16) とタマネギ (A. cepa L., 2 n = 2 x = 16) と の 種 間 体 細 胞 雑 種 を 育 成 し た . ネ ギ の プ ロ ト プ ラ ス ト か ら の コ ロ ニ ー 形 成 を 臨 害 す る た め の ヨ ー ドアセトアミド (IOA)濃 度 を 明 ら か に し た . IOA処 理 を 行 っ た ネ ギ プ ロ ト プ ラ ス ト と 分 裂 能 を 持 た な い タ マ ネ ギ プ ロ ト プ ラ ス ト を 混 合 し , ヘ テ ロ カ リ オ ン の み が 選 択 的 に 発 達 す る た め の 細 胞 融 合 法 を 構 築 し た . そ の‑115‑
結 果 , 培 養 45日目には 417個 の コ ロ ニ ー が 得 ら れ , そ の 内 の 約 80%が カ ル ス へ と 発 達 し た . 325個 中 の 33偲(10.1%)のカノレスからシユ}ト の 発 達 が 認 め ら れ た . こ れ ら の 個 体 の 中 に は 奇 形 を 示 す も の も あ っ た が , 2個 体 は ガ ラ ス 温 室 で 生 育 し た . 細 胞 遺 伝 学 的 お よ び DNA解 析 を 行 っ た 結 果 , ガ ラ ス 温 室 内 の 2個 体 は そ れ ぞ れ ネ ギ と タ マ ネ ギ の 葉 緑 体 を 有 す る 複 二 倍 体(2n

32) の 体 細 胞 雑 種 で あ る こ と が 明 ら か と な っ た . さらに,他の 3個 体 は ネ ギ の 核 ゲ ノ ム と タ マ ネ ギ 由 来 の 葉 緑 体 を 有 し て い た . こ れ ら の 個 体 の ネ ギ の 育 種 上 で の 利 用 に つ い て 考 察 し た .‑116‑
Summary
A local variety of Japanese bunching onion (Allium nstulosum L., 2 n = 2x
=16),Hakushu・negi'belonging to the variety group Senju', has been selected for taste by mass selection and cultivated in Tottori Prefecture. There remain, however, problems of susceptibility to rust disease caused by Puccinia allii and a lack of uniformity in growth due to open pollination. To resolve the former problem, resistant genes for rust disease should be introgressed, though these genes are not exist in the varieties of J apanese bunching onion. On the other hand, bulb onion (A. cepa L., 2 n = 2 x =16) is comparatively resistant to this disease (Jennings et a ,.l 1990) and these genes can be introgressed by somatic hybridization. To dissolve the latter problem, alloplasmic male sterile strains are useful to develop the F1 hybrid in Japanese bunching onion (Yamashita et al., 1999). The introduction of nuclear and/or cytoplasmic genes企omrelated species should be possible with the technology of somatic hybridization (Karim and Adachi, 1996).
In this respect, this study focused on the development of the somatic hybrid plants between Japanese bunching onion and bulb onion. To do this, the biotechnological methods, such as cell suspension culture, protoplast culture and subsequent plant regeneration, and electric protoplast fusion systems were established .
1. Establishment of cell suspension cultures and plant regeneration in Japanese bunching onion.
Callus induction for Japanese bunching onion was established on BDS medium (Dunstan and Short, 1977) supplemented with 0.2 M sucrose, 4.5
μ M 2,4‑dichloropheno勾Taceticacid(2,4・D),0.44 μ M 6・benzylaminopurine (BAP) , and 1.0 g
・
1‑1 casamino acids. Stem disc of plantlet or apical meristem region of mature plant was available as an explant of callus円t

induction. In practical use, stem disc was useful because it could be utilized all the year round in seed‑producing varieties of Japanese bunching onion. To establish cell suspension culture in J apanese bunching onion,長ltration of cell clumps by use of stainless steel sieves with 1 m m pore size was essential. It resulted that BDS medium supplemented with 200 mg ‑1‑1 casamino acids was suitable for cell suspension culture. It also revealed that higher proliferation rate ofthe cytoplasm‑rich and non‑vacuolated cells was achieved in the medium supplemented with 0.2 M sucrose. The highest rate of plant regeneration from callus was obtained on the medium supplemented with 0.5μM α‑naphthaleneacetic acid (NAA.) and 1.0 μ M BAP. The addition of casamino acids was effective for plant regeneration. It revealed that plant regeneration was capable within the 7th subculture at the rate of around 70 %, however, at the 14th subculture plant regeneration was not observed.
2̲ Establishment of protoplast culture and plant regeneration in Japanese bunching onion.
Several conditions were examined for isolation and culture of protoplasts. For isolation of protoplasts, the optimal concentration of mannitol in the enzyme solution was 0.60 M. In this condition, 9.6 x 106 protoplasts

19 FW‑1 was yielded and the viability of these protoplasts were above 95 % by the test of fluorescent diacetate (Larkin, 1976). For the isolation and purification of protoplasts by centrifuging, the force condition of 60 x g was suitable.Based on BDS inorganic salts, nutrient media suitable for protoplast culture were investigated. The optimum cell division was achieved with 5 m M potassium nitrate and a combination of 2μM 2,4‑D and 0.2 or 1μM BAP. The suitable osmolality for protoplast culture was around 0.60 osmol‑kg ・1,whereas a combination of 0.2 M sucrose, 0.2 M glucose and 0.05

M fructose accelerated cell division. It resulted that TM ‑2 (Shahin, 1985) was most e質的tiveamong the three vitamin‑types tested. Furthermore, it also revealed that suitable concentrations of inositol, ascorbic acid and cysteine was 0.56 m M, 1 m M, and 1mM, respectively. 1n addition, illumination for protoplast culture was deleterious.
The protoplasts developed into colonies by gradual reduction of sugar concentration. After 45 days of culture, numerous micro‑calli formed which were subsequently transferred to a callus formation medium. Plantlets were regenerated by transferring protoplast‑derived calli of ca. 2 m m in diameter to a modified Murashige‑Skoog medium. Ofthe 75 calli inoculated, 24 (32%) gave rise to green shoots on a regeneration medium 2 to 3 months after inoculation. These shoots rooted when cultured on a hormone合 eemedium.
These plantlets were successfully acclimatized and grew normally in a greenhouse.
3. Conditions of electric pro初plastfusion between Japanese bunching onion and bulb onion.
For the purpose of developing somatic hybrid plants between J apanese bunching onion and bulb onion, effective and reproducible conditions for electric protoplast fusion were examined. Comparative studies of various combinations on composition of fusion solution, electric parameters for fusion and protoplast density were carried out and suitable conditions could be clarified. White protoplasts of Japanese bunching onion were isolated 企omsuspension cultured cells and green protoplasts of bulb onion were from young leaves with chloroplasts. These were fused each other and the heterokaryons were distinguished from others. The 0.5 m M calcium solution was most suitable for electrofusion. The optimum electric conditions for protoplast fusion were identified; 1.50 kV • cm ‑1 for pulse五eldstrength, 40 seconds for AC time, and 50μsecond for pulse width. The highest yield of

4. Production of somatic hybrid plants between Japanese bunching onion and bulb onion via electrofusion
Interspecific so血atichybrids between J apanese bunching onion (Allium fistulosum L., 2 n
=
2 x= 16) and bulb onion (A. cepa L., 2 n=
2 x= 16) were produced by the electorofusion of protoplasts. A concentration of iodoacetamide (IOA) suitable for inhibiting the formation of colonies企om protoplasts in Japanese bunching onion was used. IOA‑treated protoplasts of J apanese bunching onion and untreated protoplasts of bulb onion were mixed together and fused, with the expectation that only heterokaryons would selectively develop into colonies. Four hundred and seventeen colonies had formed from fusion products after 45 days culture and approximately 80 % of them grew into calli. Plant regeneration was achieved for 33 out of 325 (10.1 %) calli. Some regenerants expressed abnormalities but two were successfully transplanted in a greenhouse. Cytogenetical and DNA analyses revealed these two regenerants to be amphidiploids (2 n=

32). Furthermore, it was shown that another three regenerants possessed the nuclear genome of Japanese bunching onion while their chloroplasts were from bulb onion. The application of these hybrids to a breeding program for J apanese bunching onion was discussed.‑120‑
