1. はじめに  ホパノイドは五環式トリテルペノイド骨格を持 つ化合物であり,主にバクテリアの細胞膜に由来 する(Ourrison et al., 1979)。バクテリアの生体内 で生成されるホパノイドの側鎖はヒドロキシル 基,アミノ基,配糖体などの官能基を持つことが 知られているが(Rohmer et al., 1992),自然界の 土壌,堆積物,石油などに検出されるホパノイド では,生物分解作用や続成作用によって官能基が 変化しているものが少なくない。ヒドロキシル基 を 1 つ持つホパノールについては海底堆積物,湖 底堆積物,ピート層などを用いた研究例があるが (Innes et al., 1997; Quirk et al., 1984; Venkatesan et al.,

1990),ヒドロキシ基を多数持つホパンポリオー ルについては主にバクテリアの培養試料を用いて 研究が進められてきた(Rohmer et al., 1984)。最 近,環境試料中のホパンポリオールの研究も行わ れるようになり,海底下数百メートルにある堆積 物中にもホパンポリオールが存在することが示さ れている(Saito and Suzuki, 2007)。本報では,海底 堆積物中に存在する各種ホパノールの GC/MS 分 析結果を報告する。 2. 試  料  分析試料として,ODP Leg 190 において四国沖 南海トラフで採取された Site 1178 の海底堆積物 試料を用いた。堆積物試料は海底下 130 m より採 取された泥質堆積物で,有機炭素濃度は 0.68%で あった。同試料についての他の分析結果は Saito and Suzuki(2007)に記載してある。船上で堆積物 試料を採取した後,実験室での分析まで試料を −20℃で保存し,分析直前に凍結乾燥し,粉末試 料を有機物分析に用いた。 3. 分析方法 3.1. ホパノールの抽出・分離  粉末試料 5 g を遠沈管に入れ,メタノール ⁄ ジ クロロメタン(3/1),メタノール ⁄ ジクロロメタ ン(1/1),ジクロロメタンの順にそれぞれ 15 分 間超音波抽出を 2 回ずつ行った。抽出物はロータ リーエバポレーターで乾固し,0.5N 水酸化ナトリ ウム・メタノール溶液を加え 80℃で 3 時間還流 しケン化を行った。その後,蒸留水を添加し,ヘ キサン ⁄ ジエチルエーテル(9/1)で抽出すること により中性フラクションを得た。得られた中性フ ラクションはロータリーエバポレーターで半乾固 し,ヘキサンに溶解させ,シリカゲルカラム(内 径 6 mm,長さ 8 cm)によってさらに 4 つのフラク ション(F1∼ F4)に分画した。各フラクションは それぞれ脂肪族炭化水素フラクション(F1;ヘキ サン,4 ml),芳香族炭化水素フラクション(F2; ヘキサン ⁄ トルエン,1/1,4 ml),ケトンフラク ション(F3;ヘキサン ⁄ 酢酸エチル,9/1,5 ml), 技術論文

極性脂質フラクション(F4;酢酸エチル ⁄ メタノー ル,1/1,4 ml)である。ホパノールは F4 の極性 脂質フラクション中に検出される。 3.2. ホパノールの誘導体化  多くのホパノールを検出し同定するためにトリ メチルシリル(TMS)化とアセチル化の二通りの 誘導体化を行い比較した。F4 フラクションを試験 管に移し,溶媒を蒸発させた後,BSTFA50 μl を添 加して TMS 化(60℃,1 時間)を行った。アセチ ル化は,F4 フラクションにピリジン ⁄ 無水酢酸(1 /1,4 ml)を添加し,50℃で 1 時間反応させて行っ た。なお,アセチル化した F4 フラクションを加 熱後,室温で一昼夜おき,未反応の官能基をなく すために,さらに TMS 化を行った。 3.3. ガスクロマトグラフィー質量分析(GC/MS)  TMS 化,またはアセチル化した F4 フラクショ ンをそれぞれ Hewlett-Packard 社製 HP6890 ガス クロマトグラフ/HP MSD 5973 質量分析に導入し て分析を行った。キャピラリーカラムは DB-5HT (Agilent 社製;内径 0.25 mm,長さ 30 m,膜厚 0.1 μm)を使用した。オーブン温度は,60℃で 2 分間 保持し,250℃まで 10℃ ⁄ 分で昇温し,250℃から 320℃まで 3℃ ⁄ 分で昇温後,320℃で 20 分間保持 した。キャリアガスとして高純度ヘリウムを用い た。イオン検出は質量範囲 m/z 50-750 で,2.14 (scan/sec)のスキャン速度で質量スペクトルを取 得した。なお,イオン源温度は 250℃,イオン化 電圧は 70eV に設定した。検出されたホパノール と同程度の保持時間を持つ炭素数 40 以上の高分 子量 n-アルカンが同じ試料中に存在しておらず, また,炭素数 40 以上の一連の n-アルカン標準試 料を入手することが困難であったため,本報では 保持指標を定めていない。 4. 結  果  同じ海洋堆積物中に含まれるホパノールを TMS化して得たホパノールトリメチルシリル エーテルとアセチル化して得たホパノールアセ テートの m/z 191 マスフラグメントグラムを Fig. 1の(a)と(b)にそれぞれ示した。同定された化 合物の名称や特性を Table 1 に,質量スペクトルと 構造式を Fig. 2 にそれぞれ示した。 4.1. ホパノールトリメチルシリルエーテル  TMS 化によって 5 種類のホパノールトリメチル シリルエーテルが十分に検出された(Fig. 1 およ び Table 1 のピーク番号 1-5)。ホパノイドに特徴 的な質量スペクトルである m/z 191 を持つ化合物 が他にも検出されたが,低濃度のため鮮明な質量 スペクトルが得られなかったので,ここでは報告 しない。ホパノールの同定は GC/MS による質量 スペクトルを文献と比較して行った。フラグメン ト b(M-15)はメチル基の離脱する[M-CH3]+,c (M-90)はトリメチルシラノール(C3H10OSi)が離 脱する[M-C3H10OSi]+,d(m/z 369)は側鎖の離脱 した A-E 環 C27H45+,e(m/z 191)は C 環の開裂に より ED 環と側鎖が離脱した AB 環 C14H23+ ,f(M-221)は AB 環の離脱した[M-C16H29]+,g(f-90)は fからトリメチルシラノールが 1 つ,h(f-180)は f からトリメチルシラノールが 2 つ離脱してできた フラグメントである。32, 35-アンヒドロホパンテ トロールはその質量スペクトルから最初はカルボ R el at iv e in te ns ity

ニル基を持つ C35ホパンケトジオールと同定され たが(Schaeffer, 1993),最近の NMR 解析から正確 な構造決定がなされている(Costantino et al., 2001; Bednarczyk et al., 2005)。ホパンテトロールのトリ メチルシリルエーテルの質量スペクトルはこれま で Sinninghé Damste et al.(2004)でのみ報告され ている。全ての質量スペクトルの解釈はまだ明ら かではないが,側鎖の開裂にともなう[M-193]+ や[M-307]+のフラグメントが特徴的である。 4.2. ホパノールアセテート  アセチル化によって 8 種類のホパノールトリア セテートが十分に検出された(Fig. 1 および Table 1のピーク番号 1 -8 )。フラグメントの解釈はトリ メチルシリルエーテルとほぼ同じであるが,誘導 体化試薬の違いに起因して,トリメチルシラノー ルの離脱の替わりに酢酸(C2H4O2)が離脱する。 ヒドロキシル基を 1 つ持つホパノールでは e(m/z 191)と f(M-221)の相対強度が 17, 21-立体異性体 により異なることが知られており,f/e 強度比は 17α(H),21β(H)異性体,17β(H),21α(H)異性 体,17β(H),21β(H)異性体の順に大きくなり,異 性体の識別に有用である(Quirk et al. 1984)。TMS 化とアセチル化では同じ 17β(H),21β(H)ホパ ノールであっても,f/e 強度比の傾向は異なる。 C32ホパンジオールのヒドロキシル基の位置とし て,30,31 位の炭素と 30,32 位の炭素が提案さ れている(Rodier et al., 1999; Watson and Farrimond, 2000)。質量スペクトルはほとんど同じなので,

同定には NMR スペクトルなどによる構造決定が 必要である。2 つの C34ホパントリオールの質量 スペクトルはほぼ同一であることから 22R と 22S のエピマーと考えられるが,Peiseler and Rohmer (1992)で報告されている 34R と 34S の異性体の可 能性も考えられる。  今回検出された海洋堆積物に含まれるホパノー ルの多くは,続成作用の過程で生成したジオホパ ノールである。生合成によって生成したバイオホ パノイドと同様に,堆積物中の各種ホパノールに も両親媒性のものや高沸点のものが多量に含まれ ていると考えられる。さらに多くのジオホパノイ ドやバイオホパノイドを分析するためには側鎖を 切断する化学的分解法や液体クロマトグラフィー 質量分析計(LC/MS)を用いた分析法が有効であ る。 謝  辞  本研究の遂行に当たり北海道大学大学院の沢田 健講師には貴重なご助言を頂きました。北海道大 学大学院の山本正伸准教授および匿名の査読者か らは有益なご指摘を頂きました。記して感謝致し ます。 引用文献
