To examine the effects of perinatal Gadolin-ium (Gd)-Based Contrast Agent (GBCA) injection on off-spring behavior in adulthood.
Pregnant Balb C mice were injected with macrocyclic GBCA (gadoterate meglumine), linear GBCA (gadodiamide), or con-trol from embryonic day 15 to 19 at 2 mmol kg body weight day. Gd concentrations of brain samples from dams (n 3) and offspring (macrocyclic, n 9; linear, n 10) collected on postpartum day (P) 28
plasma-mass spectrometry. Behavioral testing of offspring (control, n 22; macrocyclic, n 23, linear, n 20) was started on P70. Gd was retained in both dam and offspring brains. Concentrations were higher in the linear group (dams, 337.77 51.46 ng g vs 63.27 9.35 ng g, p 0.001; male offspring, 219.79 101.79 ng g vs 8.648 2.18 ng g; p 0.05; female offspring, 618.10 29.59 ng g vs 49.41 17.56 ng g; p 0.01). Offspring of mice receiving perinatal GBCA injections tended to show anxiety-like behavior, hypersensitivity, and impaired motor coordination and memory function. Perinatal Gd was transferred to pups and retained postnatally, possibly impair-ing brain development.
