Materials and Methods Plant m a t e r i a l
The s e e d s o f t i m o t h y (Phleum p r a t e n s e L . c v . Climax) ( 4 0 0 s e e d s / m ' ) were sown i n t h e E x p e r i m e n t a l Farm o f O b i h i r o U n i v e r s i t y i n e a r l y May i n 1 9 7 5 . Each o f 5 kg o f N , P , O , and K , O was f e r t i l i z e d t o t h e e x p e r i m e n t a l p l o t ( 1 0 a ) i n e a r l y May i n 1975 and 1976 The s t e m s o f t i m o t h y were c u t a t a b o u t 10 cm above s o i l s u r f a c e on 2 0 t h o f J u l y i n 1 9 7 6 . N o n ‑ d i s e a s e d haplocorms were c o l l e c t e d and washed with t a p w a t e r .
Prepa
γ' a t i o n o[ crude enzyme s o l u t i o n
Fourty grams o f haplocorms were c u t i n t o s l i c e s ( a b o u t 1 m m i n t h i c k n e s s ) with a s c a l p e l . The s l i c e s were homogenized i n 160 ml o f p h o s p h a t e bu 旺 e r(M/90 , pH 6 . 8 ) f o r 5 min u s i n g a m i x e r . The homogenate was s q u e e z e d through gauze , f o l l o w e d by c e n t r i f u g a . t i o n a t 10 , 000 g a t 5
