Summary
S u c c e s s f u l d e l i v e r y o f f o r e i g n g e n e s t o i n t a c t c a l l u s e s o f monocot orchardgrass (D α c t y l i s glomen α
臼L . ) and timothy (Phleum p r a t e n s e L.) was demonstrated by u s i n g an improved p a r t i c l e g u n . T h i s b i o l i s t i c d e v i c e was equipped w i t h a p o l y a c ‑ e t a l m a c r o p r o j e c t i l e w i t h GUS ‑gene c o a t e d g o l d p a r t i c l e s , d r i v e n by gas p r e s s u r e from a helium c y l i n d e r c o n t r o l l e d by a s o l e n o i d v a l v e . The p a r t i c l e g u n i s a w i d e d l y u s e f u l t r a n s f o r m a t i o n method c a p a b l e o f overcoming t h e h o s t ‑range r e s t r i c t i o n s o f Agrob α c t e r i u m and t h e r e g e n e r a t i o n problems o f p r o t p l a s t t r a n s f o r m a t i o n .

Agrob α ctenum , d i r e c t DNA t r a n s f e r (PEG / calcium p h o s p h a t e o r e l e c t r o p o l a t i o n ) , and m i c r o i n j e c t i o n a r e u s e f u l f o r t h e i n t r o d u c t i o n o f f o r e i g n DNA i n t o p l a n t c e l l s i n some p l a n t s However , t h e y a r e n o t g e n e r a l l y u s e f u l b e c a u s e o f v a r i o u s b a r r i e r R T h e s e b a r r i e r s i n c l u d e l i m i t ‑ e d h o s t range , low e f f i c i e n c y and d i f f i c u l t y i n r e g e n e r a t i n g p l a n t s from t h e t r a n s f o r m e d t i s s u e s . The u s e o f m i c r o p a r t i c l e s a s DNA c a r r i e r s might overcome t h e s e o b s t a c l e s .
