卵抽出液 (ULSS) に1/20容量の6 mM CaCl2を添加し23℃で15分間インキュベート することで間期抽出液とした。この間期抽出液と1/40容量のe.r.、1/40容量のCHX、
精子核DNA (終濃度1,000本/Pl) を混和し、23℃で35分間インキュベートした。この
反応液にp21を加えたものを、あらかじめ各種組換えタンパク質やcaffeine (終濃度 5 mM)、aphidicolin (終濃度 40 Pg/ml) を入れておいたエッペンドルフチューブに加え全 量を36 Plにした。
この反応液をよく混和してからさらに30分間インキュベートし、反応後intact nuclei isolate buffer 300 Plを加えて転倒混和しswing rotorで遠心分離した (5,000 g、4℃、5 min)。半分ほど上清を取り除いた後、さらにintact nuclei isolate bufferを500 Pl加えswing rotorで遠心分離した (5,000 g、4℃、5 min)。10 Pl程度残して上清を除き、沈殿物 に5 sample bufferを加えて混和し、100℃で5分間加熱してwestern blotting用サンプ ルとした。western blottingは4.3と同様の方法でおこなったが、一次抗体に関しては4℃
で終夜反応させた。

卵抽出液 (ULSS) に1/20容量の6 mM CaCl2を添加し23℃で15分間インキュベート することで間期抽出液とした。この間期抽出液と1/40容量のe.r.、1/40容量のCHX、
biotin-dUTPを混和し、あらかじめ各種組換えタンパク質や caffeine、精子核 DNA (終
濃度1,000本/Pl) を入れておいたエッペンドルフチューブに加え全量を5 Plにした。
この反応液をよく混和してから23℃でインキュベートし、反応後1サンプル当たり 140 PlのStop Nを加えてDNA複製反応を停止した。この時同時に20 mg/ml Proteinase K溶液 (終濃度0.2 mg/ml) を加え37℃で60分間インキュベートすることでタンパク 質を分解させた。エタノール沈殿によりDNAを回収した後、1.2 alkali loading buffer 10 Plに再懸濁し、アルカリアガロース用サンプルとした。
アルカリアガロース用サンプルをalkali agarose gelにアプライし、13 v/cm、4℃で終 夜電気泳動した。電気泳動後、ウェルをlow melting agaroseで埋め、0.25 M HClに浸 し室温で20分間振盪することでDNAを切断した。ゲルを純水で洗浄後、denaturation
solutionに浸し室温でさらに 20分間振盪した。その後、バキュームブロッターを用い
て0.4 M NaOH中でHybond-N+ membraneにDNAを転写した (50 mbar、1 hr)。
転写後 membraneを2 SSCに浸し室温で10 分間振盪した後よく乾燥させ、UVで
DNAをcrosslinkさせた。その後、blocking bufferに浸し、室温で15分間振盪すること でブロッキングをおこなった。blocking bufferにStabilized streptavidinを加えさらに15 分間振盪することでDNA中に取り込まれたbiotin-dUTP と結合させた。wash bufferで

洗浄後、substrate equilibration bufferに浸し室温で5分間振盪した。
化学発光試薬を用いてX線フィルムに造影し、現像液、停止液、定着液の順に浸し て現像した。
