放射線は酸化ストレスの一種であり、ほ乳類細胞において DNA、タンパク質、脂質への損傷、さ らにミトコンドリア依存性の活性酸素種産生の原因となり、細胞死、細胞周期停止など様々なスト レス応答を引き起こす。ヒトへの放射線影響では、代表的な疾患として急性骨髄性白血病を引き起 こすことが知られているが、その詳細なメカニズムは、未だに明らかになっていない。これらのメ カニズムの解明には、造血幹/前駆細胞に対する放射線影響を詳細に解析する必要がある。そこで 本総説では、放射線による造血幹/前駆細胞の動態変化、特に生存率/クローン増殖能、細胞周期分 布、活性酸素種、DNA 損傷/修復、染色体異常、老化について焦点をあて、最近の知見を集約した。

Ionizing radiation (IR) brings oxidative stress, and can cause damages not only on DNA but also proteins and lipids in mammalian cells, and increases the mitochondria-dependent generation of reactive oxygen species (ROS), with the subsequent induction of cell death, cell cycle arrest, and stress related responses. It is well known that IR induces acute myeloid leukemia that originates in hematopoietic cells. However, the mechanisms
