Naturalproductsof microbial origin are often biosynthesizedby multifunctionat

megasynthetases whose genetic engineering and heterolegousexpression offers

considerable promise,especially ifthenatural hostsare geneticallydifficulttohandle, slow growing, unculturable er even Lmknown

(1,2).

We have describeda

straightforward strategy that combines the power of advanced DNA engineering

(recombiogenic

cloning) inEscherichiacoli with theutility of pseudomonadsas the

heterologoushostfbrtheanalysis and mutagenesis of known and unknown secondary metabolite pathways

(2,3).

As a

first

example, themyxochromide S biosyntheticgene

cluster fromstigmatellaaurantiaca was rebuilt and engineered inE.coli to contain the

elements required forexpression in pseudomonads,The successfu1 productionin

Pseudomonas putido,at unprecedented levels,demonstratesthe feasibilityof a new

approach tothe analysis and mutagenesis of theseimportantpathways

(4).

Recently,

R puticlawas also used to express secondary metabolic genesof unknown function

resulting intheproductionofa novel compound frommyxobacteria

(5).
