heterologous
expression of myxobacterial natural

Naturalproductsof microbial origin are often biosynthesizedby multifunctionat
megasynthetases whose genetic engineering and heterolegousexpression offers
considerable promise,especially ifthenatural hostsare geneticallydifficulttohandle, slow growing, unculturable er even Lmknown
(1,2).
We have describedastraightforward strategy that combines the power of advanced DNA engineering
(recombiogenic
cloning) inEscherichiacoli with theutility of pseudomonadsas theheterologoushostfbrtheanalysis and mutagenesis of known and unknown secondary metabolite pathways
(2,3).
As afirst
example, themyxochromide S biosyntheticgenecluster fromstigmatellaaurantiaca was rebuilt and engineered inE.coli to contain the
elements required forexpression in pseudomonads,The successfu1 productionin
Pseudomonas putido,at unprecedented levels,demonstratesthe feasibilityof a new
approach tothe analysis and mutagenesis of theseimportantpathways
(4).
Recently,R puticlawas also used to express secondary metabolic genesof unknown function
resulting intheproductionofa novel compound frommyxobacteria
(5).
Inthepresentation,
further
developments
such as metabolic pathwayengineering ofRputiclato enable productionof precursorsrequired
for
secondary metabolitebiosynthesiswill bedescribed.Inaddition, applications ofthe Red/ET technologyand
datarelated tothe use of otherheterologoushostswillbepresented. References:
